First question - well I can't answer for certain but the differences seem minor to me. I would think good chain maintenance is likely to have a bigger effect on chain life.
Second question - standard =16:41 .... so 15:41 requires 1 link less and 16:44 requires 3 links more than standard. You have a bit of room to play with in the swingarm adjustment and I know that 15:41 can be accommodated on Falco and Mille swingers with a standard chain.
I'd guess that you'd probably need a longer chain to cope with the 16:44 ... but I've never tried that combo so I don't now for certain.

As I commented earlier, you need a longer Mille chain to cope with a
16 : 42, just one extra tooth on the back!

While noting all the above comments I would just say this...
The simplest and cheapest gearing mod. is to drop down to a 15 t front sprocket.
I did this almost as soon as I got mine and have done over 10k and a trip to Europe with no problems (Touch wood!) The chain did not need shortening and has shown no obvious signs of increased wear. In fact I believe it to be the original chain and rear sprocket. (17k)
It is unavoidable that there will be increased wear on the front sprocket due to there being fewer teeth to share the load but once again nothing obvious.
Give it a go - it seems to be a fairly universal early mod according to the forums.

It looks like a 15t front will do as I have a new 41t rear that came with the bike.
